I and a friend will be in Vegas next week, arriving on Thursday. During a previous trip, I earned the 500 points at Wynn so should have a free buffet available to me--at least I hope so. My question is, can this be used on Friday and/or Saturday or is it only Sunday thru Thursday?

When I earned my free buffets, the Players Club boothling
specifically told me that the free buffet was a "use it or lose it"
promotion and that I could not get the free buffet on my next
trip, unless I earned the 500 points all over again.
The guy may have been wrong, but this was MY experience.
BTW, I would never pay for the Wynn buffet. Although it was not
terrible, it was no better than many other buffets, which are a
third of the price.

Your mileage may vary, but I have successfully used a 500-point
buffet two months after earning it. Like you, I was under the
impression that I couldn't do so and that I'd lost the buffet for
good, but upon checking with the boothling they said it was still
there two months later.
