I played the 9/6 JOB spin poker at MC on Wednesday of this week and started out doing OK on it, I was at least staying even on my $100 in. Then I lost like 8 out of 10 spins in a row, betting 45 coins, a $100 goes quick, but normally when you hold a pair you can make out with a higher payback then the 45 that you are betting.
I then switched to DBL, DBL spin poker and instead of hitting bet MAX, I only bet 3 lines of 5 coins each and on the first hand to come out was dealt 4 to the royal and I got it, on the middle hand which was the 4th card position in, that completed my royal. I won $1000 instead of $3000... If I would have been betting lines 4 & 5, which paid in the top and bottom V pattern, I would have picked up an extra $2000. I was still happy, but it's the woulda, coulda, shoulda thing.
I normally always bet MAX, and going into that game for some reason I hit deal and not max and got the royal on the first hand. That was the 2nd royal that I got that week, I got one at Sam's Town and it also was on the VERY FIRST hand going into a game from the menu, which was a big surprise also, as I only held 3 cards to it, then I got the wild deuces and then aces w/kicker for $500 in 30 minutes all on the same full pay machines, at Sam's Town in Las Vegas....

Spin Poker has the exact same payback and odds as single-line, with higher volatility.
So the answer to your question is yes. You can play one line and still get full-pay as long as you play 5 coins on that line. Any additional lines you play should also be 5 coins.
Coincidentally, this particular game is available at MCC. I sometimes play single-line ($1.25/spin), and sometimes 9 lines ($11.25/spin). I got four to the royal last time I played 9, but alas couldn't hit on any of the three squares, which would have been a nice $3,000 payout.
