Many (most) casinos welcome markers for machine players. They are
best used to fund a trip bankroll in an amount for which you're
uncomfortable carrying cash. It's not a particularly prudent source
from which to fund losses that run above what you brought to the
casino for most players (although preferable to tapping an ATM if that
entails excessive fees).
Early on in my play I did use a $500 line (usually the minimum amount
available) for my quarter play. However, over time I found that I
best managed my overall bankroll when I brought to the casino only the
funds that I was prepared to lose and quit dead cold when those were
exhausted.
The mechanics of a line is that you draw markers as needed during your
visit. You then typically have 10 days in which to redeem the markers
in cash or they'll be run through as checks against the checking
account you've specified at the time of your application. (A credit
report will be pulled in considering your application to determine
your reliability for repayment and to assess the appropriateness of
your requested line amount. They may, in some cases, confirm a
current bank balance.)

With regard to concerns about "carrying too much cash". Players
should be aware that the casino can function as a short-term bank.
Most casinos will set up a "front money" or "safe keeping" account at
your request. I often use one for an extended trip, rather than keep
cash in my room safe. I'll also deposit a large win and hold it over
to future trips (I'm not particularly concerned about losing the 1%
interest I'd receive in a short-term deposit account ;). You can draw
or add funds as needed.
