YOU SHOULD, even without a room. That way future offers aren't affected.
This is true at most, but not all casinos (my personal experience to the contrary is a riverboat hotel that insists that the person receiving the comp room must check in before anyone else can get a key, even if they arrive first and their name is included on the reservation). At the other end, some casinos will extend you (based on your play) not only comp for your friend to stay on days when you're not there, but may even comp a second room - it depends BOTH on casino policy AND on how valued a customer you are to that specific casino.
In the end, ask the host to "fix" things the way you want them, explaining the circumstances for you asking for this special treatment, and they will let you know if they can do it for you or not.
Also, I have a friend who was given a comp room at the LV Hilton for a five or six day trip, and a second room for a friend with two nights comped and the remainder at casino rate. When he checked in, they said that they never comp a room for that many days, and that his friend's room would be a full rate too. After some argument, he got a compromise that was NOT satisfactory. On a subsequent trip (yes, he went there again, because he doesn't go to Vegas that much and LVH was where his offers were coming from), he therefore asked for the agreed comp and reduced rates to be confirmed in writing / by email - and it took him three or four phone calls before this was actually done - even though, with each call, he was told it would be done "right away".
So, based on that, you may wish to get not only a confirmation number, but the details of your "comp" in writing. "Real" (non-casino) hotels do this all the time, and casino-hotels should be able to comply. If they can't, beware.
